Personal Profile

Stage Ⅰ (before graduation): emphasis on fundamental researchMy research experience began with my PhD project, during which I studied theexpression and function of haloarchaeal hsp70 gene. The nucleotides flanking the startcodon in hsp70 mRNAs with very short 5'-UTRs were mutated, and theircorresponding influences on gene expression (including transcript, translation leveland translational efficiency) were analyzed. Moreover, three genes (grpE, hsp70 andhsp40) of the Hsp70 chaperone machine in Hfx. volcanii were cloned, deleted andwell characterized to analyze their function on the survival and growth of haloarchaeaunder the conditions of high temperature and low salinity. 

Stage Ⅱ (after work): emphasis on applied research, especially microbialconversion of oilcrops-based agricultural byproducts to producehigh-value-added metabolites. Following my PhD I moved to Oil Crops Research Institute of Chinese Academy ofAgricultural Science, to work with Professor Fenghong Huang on metabolism ofmicrobial oils, polyunsaturated fatty acids and iturin A, and microbial conversion ofoilcrops-based agricultural byproduct. During this period, several important genes andenzymes were identified and analyzed. In addition, recombinant bacteria andoleaginous yeasts were constructed and demonstrated to produce high levels of fattyacids. Furthermore, agricultural byproducts including rapeseed meal, soybean mealpeanut meal, sesame meal, were assessed. Rapeseed meal fermentation model foriturin A production by Bacillus amyloliquefaciens was established, and the specificroles of different rapeseed meal components on iturin A production were analyzed. Many promising strategies were attempted to achieve full utilization of rapeseed mealfor the production of high-value-added metabolites. 

StageIII (as a visiting scholar in Imperial College London): combination offundamental and applied research by Synthetic Biology and Synthetic MicrobialCommunities. This work is aiming to produce those valuable chemicals sustainably by creatingcollaborative communities of yeast with marine cyanobacteria – a microbe growingfrom CO2 and sunlight in seawater or wastewater. A cyanobacterium Synechococcuswas engineered for the production and secretion of sucrose at first. This opensavenues for sugar production in remote places from seawater or wastewater. Afterengineering Yarrowia lipolytica for sucrose consumption and optimising the mediacomposition and salinity, it was shown that the growth of Y. lipolytica can besupported by the sucrose produced by Synechococcus as the sole carbon source. Different yeast producers (including nootkatone, β-caryophyllene, parthenolide andcarotene) can be introduced into the community with cyanobacteria allowing forgrowth and potentially bioproduction from sunlight and CO2, even in placesunsuitable for traditional agriculture thus not competing for agricultural land withfood and feed sources.
